Where do we enter the $300.00 cash donation if using standard deduction.

300/600 went away. It is not available for 2022.

Is this a 2021 tax return? Is it a qualifying charity for the non-itemized deduction?

You can enter it on Line 12b of the 1040 Wks. Or you can enter it on Schedule A, and the 1040 Wks will give you an error that will make you enter it.
